This study reveals that tumor budding-like structures are common in chordomas and are closely associated with poor prognosis. Through multi-omics analysis and large-scale cohort validation, researchers identified a tumor budding-like cell subpopulation and uncovered its interaction with cholesterol metabolism tumor-associated macrophages. Mechanistically, BACH1-driven cholesterol metabolism tumor-associated macrophages secrete ANGPTL4, which activates tumor budding-like cells through SDC4, promoting their stemness, cholesterol accumulation, and malignant progression. In terms of treatment, inhibiting cholesterol metabolism or blocking the BACH1\ANGPTL4\SDC4 axis can suppress tumor invasiveness in preclinical models. Overall, this work redefines tumor budding-like structures as a key prognostic and mechanistic feature of chordomas and identifies a new immunometabolic signaling axis, providing potential therapeutic targets. |
