Urachal cancer (UrC) is a rare, aggressive malignancy typically diagnosed at advanced stages, but cytotoxic chemotherapy offers limited efficacy, and prospective clinical trials are exceedingly difficult due to the rarity of the disease. Organoids were generated from fresh UrC tumor samples collected from patients undergoing en bloc resection, profiling with high-throughput whole-exome sequencing, RNA-seq and drug assay. UrC organoids recapitulate the phenotypic and molecular features of their parental tumors, and revealed resistance mechanism and new treatment strategy. |
