Untargeted metabolomic analysis was performed using gas chromatography-mass spectrometry (GC-MS) and liquid chromatography-mass spectrometry (LC-MS) to systematically characterize the metabolic profiles of pepper cultivar Hong Guan 6 from three origins. Quality control (QC) samples were prepared by pooling equal volumes of homogenates from each pepper sample to evaluate the stability, reproducibility, and reliability of the analytical system throughout the experiment. Detailed chromatographic and mass spectrometric conditions, consistent with the study s actual operations, are described as follows:1.Gas Chromatography-Mass Spectrometry (GC-MS) Analysis1.1 Instrument ConfigurationGas chromatograph: Agilent 7890B (Agilent Technologies Inc., Santa Clara, CA, USA)Mass spectrometer: Agilent 5977B MSD (Agilent Technologies Inc., Santa Clara, CA, USA)Chromatographic column: DB-5MS fused-silica capillary column 1.2 Chromatographic ConditionsSample preparation: Derivatization was performed based on the LC-MS extracted supernatant. 150 μL of the LC-MS supernatant was transferred to a glass vial and completely dried using a centrifugal vacuum concentrator. The oximation reaction was carried out by adding 80 μL of methoxyamine hydrochloride (15 mg/mL in pyridine) and incubating at 37°C for 60 min with shaking. Subsequently, a derivatization cocktail consisting of 50 μL N,O-bis(trimethylsilyl)trifluoroacetamide (BSTFA), 20 μL n-hexane, and 10 μL of internal standard mixture (C8-C24 FAMEs in chloroform) was added, and the reaction proceeded at 70°C for 60 min. After cooling to room temperature for 30 min, the derivatized samples were analyzed. |
