The common wheat line NC99BGTAG17 carries the Pm37 gene, which confers ultra-broad-spectrum resistance to powdery mildew. Our initial attempts to clone this resistant allele failed because it is absent from the Chinese Spring reference genome. To overcome this, we used PacBio HiFi sequencing to generate a contig-level assembly of NC99BGTAG17. This produced a 17.9 Mb contig spanning the entire Pm37 physical mapping interval, which enabled us to clone the full-length sequence of NLR-1. Finally, the assembly contigs were scaffolded using the IWGSC Chinese Spring RefSeq v2.1 reference genome. |
