To investigate the pathogenic mechanism of NOTCH2NLC GGC repeat expansions in NIID, we established induced pluripotent stem cell (iPSC) lines derived from skin fibroblasts of three NIID patients carrying the mutation and three matched healthy controls. These iPSC lines were subsequently differentiated into brain organoids. At day 90 of culture, the three biological replicates from each group were pooled evenly to construct a single sequencing library for single-cell RNA sequencing. |
