Acinetobacter baumannii is a multidrug-resistant nosocomial pathogen causing infections that are difficult to treat with conventional antibiotics. Considering this challenge, bacteriocins are being regarded as complementary approaches to antibiotics. Here, we have shown that two bacteriocins produced by Gram-positive bacteria inhibit the growth of multidrug-resistant clinical isolates of A. baumannii, albeit at high concentrations. In vitro characterization revealed that they permeabilize phospholipidic vesicles, suggesting their potential to enhance the efficacy of antibiotics disrupting cell envelope integrity. Low doses of these bacteriocins potentiate low doses of colistin or imipenem, two antibiotics disrupting the cell envelope by different mechanisms, thereby enabling a substantial reduction in survival by compromising the inner membrane. Importantly, these combinations hinder both colistin tolerance and emergence of colistin-resistant mutants, which are rapidly selected during colistin treatment, and partially resensitizes a highly colistin-resistant clinical isolate. These findings illustrate the efficacy of rationally designed combination treatments involving antibiotics and bacteriocins. |
