Description |
C.variabilis was purified via coating on solid BG11 medium and then expanding propagating (maintain the initial optical value at 0.05, about 1x105 cell/mL) to collected samples by centrifugation.Three grown conditions including 400ppm (CK), long-term acidification under 1000ppm (LT) and short-time acidification under 1000ppm (ST) were carried out prior to extract preparation (The long-term acidification lasted for 680 days at 1000ppm. The short-term acidification through transferred from ambient CO2 concentration to 1000ppm about 20 generations).Separation of mRNA from total RNA was carried out by magnetic breads with oligo (dt). The mRNA was fragmented in the presence of fragment buss followed by end repair. |
