Male GF Sprague-Dawley (SD) rats (male) were obtained from the Department of Laboratory Animal Science, Peking University Health Science Center, Beijing, China. GF rats were divided into three groups and housed in separated sterilized isolators. At six weeks of age, GF rats in different groups received faecal microbiota transplantation [obtained from one IBS patient (GI group) and one healthy control (GH group) and sterile saline (GN group)] separately by gavage. Rats in the same group received faecal microbiota from the same sample, and transplantation was carried out only once. Two weeks after FMT, faecal samples of rats were collected for further microbiota analyses. Colorectal distension (CRD) tests were carried out 24 days later to FMT as described previously [44]. Then, GF rats were sacrificed with anaesthesia. Colon tissue was collected and fixed in 10% buffered formalin for immunohistochemistry. |
