Single-cell RNA five-end-sequencing methods can simultaneously quantify gene expression and detect transcribed cis-regulatory elements (tCREs, mostly promoters and enhancers) in a single assay with no extra cost, which are critical for studies of gene regulation. To enable sample multiplexing and cost-effective single-cell RNA five-end-sequencing for millions of individual cells, here we develop an efficient, simple and flexible 'Five-prime end single-cell combinatorial indexing RNA-Seq (Fipresci-Seq)' approach. In this study, we benchmarked Fipresci-Seq on an interspecies mixture of human and mouse cells. |
