Aliqouts of 50,000 cells were combined with washed Concanavalin A coated magnetic beads (BioMagPlus, BP531), and incubated with specific antibody in digitonin condition. Then, cells were incubated and fragmented by recombinant Protein-A Tn5 transposome. The tagmented DNA was collected and purified for setting up DNA libraries. |
