KIAA1199-overexpressing and vector control colorectal cancer cells were orthotopically implanted into the colorectum of BALB/c mice. After 14 days, livers including tumor-associated regions were harvested and processed into single-cell suspensions by mechanical and enzymatic dissociation. Single-cell capture, barcoding, and cDNA library construction were performed on the BD Rhapsody single-cell analysis system following the manufacturer protocol. The resulting libraries were subjected to high-throughput sequencing to profile the hepatic immune microenvironment. |
