概要: RNA-seq analyses of drought-treated rice leaves were performed to screen long noncoding RNAs showing differential expression in a time-dependent manner.
项目整体设计: Eight transcriptome profiles of drought-treated 5-week-old rice leaves were generated by deep sequencing using Illumina HiSeq 4000.
Rice seeds were germinated on MS media at 28℃ for 4 days, and transplanted into soil pots (4x4x6 cm; 3 plants per pot) and grown in a greenhouse.
处理方案:
Watering was withheld at 5 weeks after transplanting to soil and the soil water content in each pot was adjusted to approximately 75%. Soil moisture was monitored during the drought treatment using a Soil Moisture Sensor SM150.

Total RNAs were extracted from rice leaves using Trizol reagent, purified with RNeasy Mini Kit and treated with DNase I.
