Summary: We examined the global transcriptome of individual cells in the three cell populations, starting MC from three donors after 1hr adherence to FN pooled, MC-derived EC subjected to shear stress, and HCAEC subjected to shear stress using single cell RNA-sequencing (scRNA-Seq). Analysis showed that the initial population of MC clustered separately from HCAEC, while the resulting MCEC overlapped with HCAEC and not MC. Co-expression analysis reveals that only the resulting MCEC contain cells co-expressing KDR and PROM1 (CD133), whereas our starting MC population contained only one cell expressing KDR and a separate cell expressing PROM1 suggesting that our initial MC population did not contain any EPC/ECFCs. Interestingly, heatmaps constructed using EC and MC associated genes revealed that the resulting MCEC cells expressed predominantly EC genes but also some MC genes.

Overall Design: Examination of global transcriptome of 3 cell populations, initial monocytes, monocyte derived endothelial cells, and human endothelial cells as a control
