Cryopreserved PBMC samples were thawed and resuspended in complete cell culture media at a concentration of 5 × 106 cell/mL and stimulated with 0.5 μg/mL of a SARS-CoV-2 Spike protein peptide pool for 18 h at 37 °C. Spike-reactive CD4+ T cells were identified by expression of the activation markers CD134 and CD69 and isolated by flow cytometric sorting using BD FACSAria II instrument.
