PRJNA799046: Transcriptome analysis of piperonylic acid-treated, Meloidogyne graminicola-inoculated rice roots

Source: NCBI / GSE194096
Submission Date: Jan 20 2022
Release Date: Dec 06 2022
Update Date: Dec 06 2022

Summary: We used mRNA sequencing to study the effects of foliar piperonylic acid treatment on the transcriptome of rice plants (cv. Nipponbare) and infection by M. graminicola to gain insights in the molecular basis of piperonylic acid-induced disease resistance against this nematode

Overall Design: mRNA sequencing of the root systems of rice seedlings treated with piperonylic acid or a mock treatment 7 and 13 days post transplantation and inoculated with M. graminicola 14 days post transplantation

Protocol
Growth Protocol: Rice seedlings (cv. Nipponbare) were germinated on wet tissue paper (28°C, dark), transplanted to individual PVC tubes 5 days after germination and placed in a growth chamber (28°C, 12 hours of light)
Treatment Protocol: Plants were foliarly sprayed with a solution containing 300 µM PA and 0.1 v/v% Tween 20 or a corresponding mock solution at two time points: 7 and 14 days post transplantation.
Extract Protocol: Plants were washed, pooled (4 plants per sample) and root and shoot tissues were ground seperately in liquid nitrogen. RNA was extracted using a Zymo Plant RNA kit as per manufacturer's instructions (Zymo Research).
Library Construction Protocol: QuantSeq 3′mRNA-Seq Library Prep Kit (Lexogen) as per manufacturer's instruction, starting from 2 µg RNA.
