Autophagy has been viewed as an adaptive survival response to various stresses, whereas in other cases, it promotes cell death. Therefore, both deficient and excessive autophagy may lead to cell death. Pharmacological modulation of autophagy has been referred to as a promising therapeutic strategy for cancer. We have shown that 0404 is probably an attractive agent for treating hepatocellular carcinoma (HCC), especially in HCC with wide type (WT) p53, through forming a p53/miR-34a/SIRT1 signal feedback loop to promote cell apoptosis.
