Single-cell transcriptomic profiling of small-cell lung cancer reveals intimate interactions between tumor cells and tumor microenvironment

Small-cell lung cancer (SCLC) is the most aggressive and lethal subtype of lung cancer for which better understandings of its biology are urgently needed. Single-cell sequencing technologies provide an opportunity to profile individual cells within the tumor microenvironment (TME) and investigate their roles in tumorigenesis processes. Here, we performed high-precision single-cell transcriptomic analysis of ~5,000 individual cells from primary tumors and matched adjacent normal lung tissues (NATs) from 11 SCLC patients, including one patient with primary and relapsed tumors. By comparing to NATs, our study revealed an immune suppressive landscape of human SCLC. Malignant cells of SCLC tumors exhibited the diversity of states mainly regarding to the cell cycle, immune and hypoxia properties. Our data also revealed intra-tumor heterogeneity of key transcriptomic factors of SCLC at single cell resolution, and uncovered related genes expression patterns and functions.
