CRISPR-Cas9 Knockout Screen Identifies factors involved in the cytotoxicity of Decitabine

Description:
Decitabine (DAC), a first-line drug for acute myeloid leukemia (AML), has been long-recognized for its DNA demethylation effects. However, its full spectrum of anti-tumor mechanisms remains incompletely understood. Here, we employ a temporally resolved genome-wide CRISPR-Cas9 screen to identify key factors governing DAC response. We also use lncRNA-seq and ChIP-seq techiques to investigate the changes of trancriptome and chromatin occupancy of proteins of interest.
