Human receptive endometrial organoid for deciphering the implantation window

Human endometrial receptivity determines the success of pregnancy; however, in vivo studies of its features and regulation are particularly challenging due to ethical restriction. Recently, developed human endometrial organoids present a powerful model to dissect this complexity. Here, we established specific human window-of-implantation (WOI) endometrial organoids that can mimic the in vivo receptive endometrium. It not only structurally consists of glands and stroma but also shows similar cell types compared with the in vivo endometrium at the single-cell level. The WOI endometrial organoid also showed hormone response, secretion function and special characteristics of the implantation window, such as decidualization, ECM remodeling, pinopode generation, angiogenesis and immunoregulation. The model also clearly resolved the process of proliferation-secretory transformation, ciliary epithelium differentiation and EMT during the implantation window in vitro.
