PBMC were isolated via Lymphocyte Separation Media (Multi Science, 70-LSM01) following the protocol. Cells were washed twice using cold PBS and total RNA was extracted with the RNeasy micro kit (QIAGEN) according to the manual. RNA sequencing was performed with a Truseq RNA library prep kit v2 (Illumina) and illumina NovaSeq 6000 equipment. We used a perlscript to filter out low quality reads and subsequently the clean reads were mapped to human genome hg19 with TopHat2. Gene expression levels were calculated and normalized to fragments per kilobase of transcript per million mapped reads (FPKM).
