RNA sequencing of PBMC from LACC patients after treatment with chemoradiotherapy

Locally Advanced Cervical Cancer (LACC) is a highly lethal malignancy affecting women. Analyzing the transcriptional changes in peripheral blood mononuclear cells (PBMC) can provide valuable insights into patients' immune systems. In this study, PBMC samples were collected from 14 LACC patients who underwent chemoradiotherapy, prior to the administration of anti-programmed cell death-1 (PD-1) antibodies. Based on the observed outcomes following the PD-1 antibody treatment, the patients were subsequently categorized into two groups: those experiencing immune-related adverse events (irAE) and those without. This project aims to utilize bulk RNA-seq technology to analyze the gene expression profiles of PBMC from LACC patients after chemoradiotherapy.
For each controlled access study, there is a corresponding Data Access Committee(DAC) to determine the access permissions. Access to actual data files is not managed by NGDC.
Radiotherapy is the first-line treatment for locally advanced cervical squamous cell cancer (CSCC), however, approximately 50% of patients progressed after radical radiotherapy and the molecular mechanisms are unclear. Thus, understanding of molecular responses of different cell types in tumor exposed to radiotherapy is pivotal for improving the efficiency of radiotherapy in CSCC, but single-cell level studies remain lacking. Here, we performed single-nucleus RNA-seq to construct high-resolution molecular landscape of various cell types in CSCC before and during radiotherapy.
