Description:   Maintaining chondrocyte homeostasis is crucial for the effective treatment of osteoarthritis (OA). Chondrocytes reside in a hypoxic environment under physiological conditions. This study examined the association between epigenetics and chondrocyte homeostasis and observed significant alterations in the histone acetylation in chondrocytes under hypoxic conditions. A key enhancer element was found to regulate CADM1-AS1, which plays a critical role in maintaining cartilage homeostasis. Further mechanistic investigations revealed that CADM1-AS1 recruits histone deacetylase complexes to suppress the transcriptional activation of NOS2, thereby affecting chondrocyte amino acid metabolism in chondrocytes. Conditional knockout of CADM1-AS1 in chondrocytes accelerated OA development in mice.
