HeLa-mt-Keima cells were treated by 20 mM HC for 16 hrs. Treated cells were trypsinized, filtered through a 40 um cell strainer and resuspended in PBS containing 2% FBS. Cell sorting was performed using a BD FACSAria II instrument with two channels: 405 nm excitation for mt-Keima at pH 7 and 562 nm excitation for mt-Keima at pH 4, with a 610 nm emission bandwidth. The top 25-30% and bottom 25-30% cells were sorted to represent mitophagy-enhanced and -inhibited cell subsets, respectively. |
