| Description | T cells were isolated from donor peripheral blood mononuclear cells (PBMCs), and genomic DNA was extracted. The obtained genomic DNA was randomly fragmented and then equally divided into two groups: one experimental group was subjected to in vitro cleavage using a pre-assembled ribonucleoprotein complex consisting of Cas9 protein and sgRNA, while the other served as an untreated wild-type control. These two sets of samples were subsequently used for AID-seq experiments. |
