We adjusted the NP-S buffer and used 100 units of MNase to digest nuclei for 5 minutes. After bound nucleosomes digested with Protease K, we exacted core particle DNA. Paired-end libraries were constructed using Illumina compatible DNA-Seq NGS library preparation kit from Gnomegen and were sequenced with Illumina HiSeq 2500 (PE125, paired-end 125 bp). |
