Total RNA was extracted using a mirVana miRNA Isolation Kit (Ambion). Small RNAs had linkers ligated to them and bar-coded cDNAs were prepared using a TruSeq Small RNA Sample Prep Kit (Illumina) following the manufacturer’s instructions. The amplified cDNA constructs were purified from agarose gel in preparation for sequencing analysis using the Illumina HiSeq 2500(Illumina). |
