Fresh gastric tumor and adjacent tissues were cut into approximately 1 mm3 pieces in RPMI-1640 medium (Invitrogen) with 10% fetal bovine serum (FBS; ScienCell) and enzymatically digested with a MACS tumor dissociation kit (Miltenyi Biotec) for 30 min on a rotor at 37°C, according to the manufacturer’s instructions. After filtration with a 70 μm Cell-Strainer (BD) in RPMI-1640 medium (Invitrogen), the suspended cells were centrifuged at 400× g for 5 min. The single cell suspensions were stained with 7-AAD Viability Staining Solution (Cat# 00-6993-50, eBiocience) for flow cytometry (FACS), performed on a BD Aria III instrument. Based on FACS analysis, 1 × 105 living cells were sorted into 1.5 ml tubes with sorting buffer, and counted manually under the microscope. Then, single cells were processed with the GemCode Single Cell Platform using the 3’ GemCode Gel Bead, Chip and Library Kits (10x Genomics) as per the manufacturer’s protocol. |
