For cultured cells, Ankrd52-null or control MC38 cells were seeded onto 6-well plates at 5 ×10^5 cells/well in duplicate and challenged with 10 ng/ml IFNgama for 24 hr, with cytokine-free medium serving as control. RNA isolation, library construction and sequencing were performed by WuXi NextCODE. RNeasy MinElute Cleanup Kit (QIAGEN, 74204) was used for RNA extraction, followed by generating mRNA-focused sequencing libraries from total RNA using Illumina TruSeq RNA Sample Preparation Kit v2. Paired-end 150 bp sequencing was done on illumina Hiseq X10 (40M reads for each sample). |
