Polyadenylated mRNA from total qualified RNA are enriched using poly-T oligo-attached magnetic beads. Add Fragmentation Buffer to the purified mRNA to make the them into short fragments, and then use the fragmented mRNA as a template to synthesize the first and second strand of cDNA. Purified by QIAQuick PCR kit and eluted with EB buffer. Then the target size fragments are enriched by agarose gel electrophoresis and PCR amplification is performed to construct the entire library. Qualified libraries were sequenced on the Illumina NovaSeq 6000. The sequencing strategy is PE150. |
