Total RNA was extracted from the roots, stems, and leaves of a single I. indigotica plant and its hairy roots using Trizol reagent (Invitrogen, USA), according to the manufacturer’s instructions. Quality control was performed using the electrophoresis system. For each sample, the mRNA was purified using oligo (dT)-attached beads and fragmented into small pieces. The cDNA libraries were constructed using a TruseqTM RNA sample prep Kit (Illumina, USA). Sequencing was performed on an Illumina HiSeq4000 platform (Illumina, USA). |
