The RNA sequencing was performed by Novogene Co. Ltd (Beijing, China). Total RNAs from specimens were extracted using TRIzol reagent (Invitrogen, Carsbad, CA). The concentration of extracted RNA was measured using the Qubit® RNA Assay Kit in a Qubit® 2.0 Fluorometer (Life Technologies, CA, USA). The integrity of RNA was determined with an RNA Nano 6000 Assay Kit of the Bioanalyzer 2100 system (Agilent Technologies, CA, USA). Then, sequencing libraries were generated using the NEBNext® UltraTM RNA library Prep Kit for Illumina® (NEB, USA). The library preparations were sequenced on an Illumina Hiseq platform. Raw data was firstly processed using in-house perl scripts. |
