The total bacterial DNA from all fecal samples were extracted using stool genomic DNA extraction kit (Tiangen Biochemical Technology, China) according to the manufacture instructions. The PCR amplicons of 16S V4 section were obtained using primers of 515F-GTGCCAGCMGCCGCGGTAA and 806R-GGACTACHVGGGTWTCTAAT and purified by Glycogel DNA Recovery Kit (Tiangen Biochemical Technology, China). The concentration of the purified PCR products were determined by Qubit3.0 fluorometer (Life Invitrogen, USA). The DNA library was constructed according to the instructions of the VAHTS Universal DNA Library Prep Kit (Vazyme, China) and sequenced by Illumina Miseq after quality check by Agilent 2100 Bioanalyzer System. |
