DNA was fragmented in a g-TUBE (Covaris), followed by end-repair, adapter ligation and exonuclease digestion following the Pacific Biosciences kit instructions. DNA fragments of 10-50 kb were recovered by Blue Pippin electrophoresis (Sage Sciences). Five libraries with 20-kb DNA inserts were constructed using a SMRTbell Template Prep Kit (PacBio). |
