Sequencing libraries were generated using NEBNext® Ultra™ RNA Library Prep Kit for Illumina® (NEB, USA) according to manufacturer’s recommendations. Index codes were added to attribute sequences to each sample. mRNA was purified from total RNA using poly-T oligo-attached mag netic beads. The first strand cDNA was synthesized using random hexamer primer and M-MuLV Reverse Transcriptase (RNase H-) while the second one was done using DNA polymerase I and RNase H.The library preparations were sequenced on an Illumina Hiseq 2000/2500 platform and 100bp/50bp single-end reads were generated. |
