The stem wrere harvested and cleaned with tap water and used for DNA extraction (Porebski et al., 1997). The quality of DNA was examined by agarose electrophoresis and on a NanoDrop 2000 photometer (NanoDrop Technologies, Wilmington, DE) and a Qubit 3.0 Fluorometer (Life Technologies, Carlsbad, CA). For next-generation sequencing (NGS), DNA libraries were constructed using a VAHTS Universsal Plus DNA Library Prep Kit for MGI (Vazyme, Nanjing, China) following the manufacturer’s instructions, and the libraries were sequenced on an MGI-SEQ 2000 (MGI Tech, Shenzhen, China) to generate 150 bp pair-end data. A SMRT bell CLR library for DNA long reads were constructed using a SMRT bell Express Template Prep kit 2.0 (Pacific Biosciences, CA), and on a PacBio Sequel II platform subreads were generated. |
