Total RNA was extracted using TRIzol reagent. mRNA was enriched usng Oligo（dT）magnetic beads, followd by random sheering in fragmentation buffer. mRNA was then used as templates to systhesize the cDNA. Ploy A tail was added to the purified cDNA, followed by segment size selection using AMpure XP beads. The library was then enriched by PCR. |
