The nucleic acid was extracted from the 140 ul anal swab. Reverse transcription (RT)-PCR was performed by using random primers. The cDNA was then converted to Second-stranded DNA and library preparation was carried out using the SQK-LSK109. Sequencing of metatranscriptome libraries was conducted on the Nanopore GridION platform and about 3,000,000 reads were generated. |
