Genomic DNA was extracted and sequenced according to the manual of Ligation Sequencing Kit (Nanopore, Oxford shire, UK). Briefly, the DNA was purified and assessed using a Qubit 2.0 Fluorometer (Thermo Fisher, CA, USA). The DNA fragments of -20 kb were enriched and purified after randomly sharing. After ends repaired and adapters ligated, a 20-kb library was constructed for the sequencing. |
