Freshly prepared chromatin was fixed with formaldehyde and extracted. Fixed chromatin was then digested with DpnII and the sticky ends were filled in with biotinylated nucleotides before ligation. Crosslinking was then reversed. DNA was purified and treated to remove biotin from unligated ends. DNA was then sheared and the biotin-containing fragments were enriched through streptavidin bead pulldown. |
