DNA was extracted from the faeces or microbiota using a QIAamp MagAttract HMW DNA Kit, we constructed metagenomics co-barcoding sequencing system, which named as MECOS. Next, we would complete the MECOS library construction use high-molecular-weight DNA from faeces. To achieve more bacteria, we combined the culturing procedures for human microbiota and MECOS which named as cMECOS. The sequencing libraries of MECOS and cMECOS were constructed based on customized process, which mainly included the following steps: 1) Transposon insertion; 2) Co-barcoding of long fragment; 3) Ligation and PCR; 112) DNB generation and sequecing. |
