The cDNA libraries were constructed with approximately 5 ug of RNA for each sample using the NEBNext Ultra RNA Library Prep Kit according to the manufacturer's instructions and index codes were added to attribute sequences to each sample. To select cDNA fragments of preferentially 250-300 bp in length, the library fragments were purified using the AMPure XP System. Then PCR was performed by Universal PCR primers, Index (X) Primer, and cDNA. The PCR products were purified and library quality was assessed on the Agilent Bioanalyzer 2100 system. Each library was used for Paired-End sequencing using Illumina HiSeq4000. |
