The macrovirulome library is built by removing ribosomal RNA from the total RNA and then building the library according to the NEB normal library building method, specifically: firstly, the ribosomal RNA is removed from the total RNA to obtain mRNA; subsequently, the obtained mRNA is randomly broken into short fragments of 250-300 bp with divalent cations in NEB Fragmentation Buffer, and the fragmented RNA is used as the template and random Oligonucleotides were used as primers to synthesize the first strand of cDNA, followed by PCR amplification according to the NEB common library building method, and finally the library was obtained on the machine. |
