The sample was fixed with formaldehyde, and DNA was enzymically cut by restriction enzyme to produce sticky ends on both sides of the crosslinking. Biotin-labeled bases were introduced, the end-repaired DNA was cyclized, DNA was decrosslinked, DNA was purified, and DNA was broken into fragments of 300 bp-700 bp. The DNA fragments containing interaction relationships were captured by the magnetic beads of strand avidin for library construction. |
