Fresh young leaf tissue was collected and immediately frozen in liquid nitrogen, and high-quality DNA was isolated from fresh young leaves using the CTAB method and assessed with the NanoDrop One spectrophotometer (NanoDrop Technologies, Wilmington, DE)). The DNA quantity was measured with a Qubit 3.0 Fluorometer (Life Technologies, Carlsbad, CA, USA). For SMRT sequencing, a standard HiFi library was prepared according to the SMRTbell Express Template Prep Kit 2.0 manual (Pacific Biosciences, CA, USA). The genome of M. notabilis was sequenced using the PacBio Sequel II system (Pacific Biosciences) |
