Fresh young leaf tissue was collected and immediately frozen in liquid nitrogen, and high-quality RNA was isolated from fresh young leaves using the CTAB method and assessed with the NanoDrop One spectrophotometer (NanoDrop Technologies, Wilmington, DE)). The RNA quantity was measured with a Qubit 3.0 Fluorometer (Life Technologies, Carlsbad, CA, USA). IsoSeq reads from the ONT platform were sequenced by NanoFilt (v2.8.0, https://github.com/wdecoster/nanofilt), Pychopper (v2.7.2, https://github.com/epi2me-labs/pychopper), Racon (v1.4.21) (Vaser et al., 2017), and minimap2 (Li, 2018) to generate .bam files.
