The CUT&RUN assay was conducted on 0.2 million cells per sample, utilizing the Hyperactive pG-MNase CUT&RUN assay kit (Vazyme, HD102) with slightly modifications to the manufacturer’s protocol. The libraries were prepared using NEBNext Ultra II DNA library prep kit (NEB, E7645) with modified amplification condition as 98 °C for 30 sec, 15 cycles of 98 °C for 10 sec and 65°C for 17 sec, final extension at 65 °C for 2 min and hold at 4°C. |
