For comparative transcriptome sequencing, the cells were cultured under modified By+ medium for 24 h, and resuspended in nitrogen-free medium afterwards and cultured under 15ºC, 20ºC, 30ºC and 35 ºC, respectively. After induction for 24 h, samples were harvested at 4ºC and rapidly frozen in liquid nitrogen. The cells were ground with the aid of liquid nitrogen, the RNAiso plus reagent. was used to extract total RNA. The concentration of RNA was measured with Nanodrop 2000. About 14 μg of RNA was used to synthesize cDNA using the cDNA synthesis kit. |
