The single cell of each specimen was isolated using a stereo-microscope combined with a micromanipulator equipped with a glass micropipette. Samples were reverse-transcribed, template-switched, amplified and cDNA libraries were built by modification of the Smart-seq2 single-cell RNAseq method. Use the Illumina sequencing platform to generate paired end reads. |
