Young and tender leaves were fixed in 2% formaldehyde solution, and the crosslinked DNA was then digested with DpnII (NEB, UK). The sticky ends of the digested DNA fragments were attached with biotin-labeled adapters, which were specifically enriched to obtain fragments of approximately 500 bp to construct the Hi-C library. |
