The total DNA was then extracted with the QIAamp DNA Mini kit (QIAGEN). The 16S rRNA high-throughput sequencing was performed by Realbio Genomics Institute (Shanghai, China) using the Illumina MiSeq. Variable regions V3-V4 on 16S rRNA genes of bacteria were amplified with forward primer F341 5’-ACTCCTACGGGRSGCAGCAG-3’ and reverse primer R806 5’-GGACTACVVGGGTATCTAATC-3’. The raw data were then subjected to a quality control procedure using UPARSE. The qualified reads were clustered to generate operational taxonomic units (OTUs) at the 97% similarity level using Usearch. |
